Lipid-derived lipoxins are produced at the site of vascular and mucosal inflammation where they down-regulate polymorphonuclear leukocyte recruitment and function. 15(R)-Lipoxin A (15(R)-LXA4) is derived from the aspirin-triggered formation of 15(R)-HETE from arachidonic acid . 15(R)-LXA inhibits LTB-induced chemotaxis, adherence, and transmigration of neutrophils with twice the potency of LXA demonstrating activity in the nM range. The anti-inflammatory effects of aspirin may be ascribed in part to the ability of 15(R)-LXA to regulate leukocyte function. 15(R)-LXA is reported to promote resolution of inflammation in LPS-treated stromal cells derived from intermediate-stage diseased supraspinatus tendons as evidenced by increased expression of the STAT-6 pathway target genes, ALOX15 and CD206.

15(R)-Lipoxin A4 is a specialized pro-resolving mediator and an acetylated derivative of COX2. 15 (R)-Lipoxin A4 is present in neurons. 15 (R)-Lipoxin A4 induces the SPM synthases ALOX12 and ALOX15, as well as the pro-resolving receptor ALX. 15 (R)-Lipoxin A4 inhibits protein kinases, including JNK1/2/3, Lyn, STAT-3 and STAT-6. 15 (R)-Lipoxin A4 enhances the release of pro-resolving mediators. 15 (R)-Lipoxin A4 alleviates the pro-inflammatory phenotype of tendon-derived stromal cells. 15 (R)-Lipoxin A4 promotes the resolution of neuroinflammation. 15 (R)-Lipoxin A4 is applicable to research related to achilles tendinitis, achilles tendon rupture and Alzheimer’s disease.

15 (R) - Lipoxin A4 (10 nM; 24 h) can reduce the level of pro-inflammatory arachidonic acid in tendon stromal cells derived from Achilles tendon disease stimulated by IL-1 β and increase the level of specific pro regressive mediators.
15 (R) - Lipoxin A4 (10 nM; 24 h) can reduce the levels of pro-inflammatory arachidonic acid in tendon stromal cells derived from Achilles tendon rupture stimulated by IL-1 β, and increase the levels of pro-inflammatory mediators .
15 (R) - Lipoxin A4 (10 nM; 24 h) can upregulate the expression of pro regressive biosynthetic enzymes ALOX12, ALOX15, and receptor ALX in tendon stromal cells derived from Achilles tendon disease and Achilles tendon rupture stimulated by IL-1 β, and the mRNA expression of ALOX15 was significantly increased in both cell types.
