Lactate (Lactate acid) is the product of glycolysis. Lactate is produced by oxygen lack in contracting skeletal muscle in vivo, and can be removed under fully aerobic conditions. Lactate can be as a hemodynamic marker in the critically ill.
体外研究

Lactic acid (20 mM; 24, 48 hours) significantly reduced cell proliferation and migration, and converted cell metabolism to oxidative phosphorylation.
Lactic acid (20 mM; 24 hours) significantly increased the protein and mRNA expression levels of MCT1 and HCAR1 .

Cell Proliferation Assay

Cell Line: Uveal melanoma 92.1 cells
Concentration: 20 mM
Incubation Time: 24, 48 hours
Result: Inhibited cell proliferation and migration.

Western Blot Analysis

Cell Line: Uveal melanoma cells
Concentration: 20 mM
Incubation Time: 24 hours
Result: Showed an increase in the protein expression levels of MCT1 and HCAR1
体内研究

Lactic acid (1.6 g/kg; pH 7.4; injection in white); Once a day; From day 8 to day 30, CD8+T cell-dependent tumor growth inhibition was observed in the MC38 colon cancer model.
Note: Before administering drugs to animals, the pH of the working solution needs to be adjusted to around 7.4.

Animal Model: Female C57BL/6 mice with MC38 colon cancer cells
Dosage: 1.6 g/kg
Administration: Subcutaneous; daily from Day 8 to 30 days
Result: Significantly suppressed tumor growth.

  • 1.

    请依序添加每种溶剂: 10% DMSO → 40% PEG300 → 5% Tween-80 → 45% saline

    Solubility: ≥ 5 mg/mL (55.51 mM); Clear solution

    此方案可获得 ≥ 5 mg/mL (55.51 mM,饱和度未知) 的澄清溶液。

    以 1 mL 工作液为例,取 100 μL 50.0 mg/mL 的澄清 DMSO 储备液加到 400 μL PEG300 中,混合均匀;向上述体系中加入50 μL Tween-80,混合均匀;然后继续加入 450 μL生理盐水定容至 1 mL。
