Ceftolozane (CXA-101) sulfate 是一种抗假单胞菌 cephalosporin。Ceftolozane sulfate 可与铜绿假单胞菌必需 PBP (1b、1c、2 和 3) 高亲和力结合从而抑制细胞壁合成。Ceftolozane sulfate 可抑制铜绿假单胞菌和肠杆菌科细菌。

生物活性

Ceftolozane (CXA-101) sulfate is an antipseudomonal cephalosporin. Ceftolozane binds to P. aeruginosa essential PBPs (1b, 1c, 2 and 3) with high affinity. Ceftolozane inhibits cell wall synthesis by binding the PBPs. Ceftolozane sulfate inhibits P. aeruginosa and Enterobacteriaceae.

体外研究

Ceftolozane sulfate exhibited antibacterial activity against Pseudomonas aeruginosa with complete AmpC inhibition, with a MIC of 4 mg/L.

Ceftolozane sulfate (≤ 0.12->64 μg/mL) exhibited anti pseudomonas activity against carbapenem resistant and MDR isolates, with MIC>8 μg/mL against 11 carbapenem resistant Pseudomonas aeruginosa isolates (4.7%).

体内研究

Ceftolozane (400-800 mg/kg, injected in white every 6 hours) can reduce the production of ESBL by Escherichia coli bacteria with neutropenia.
Ceftolozane (180 mg/kg, subcutaneous injection, once every 8 hours, 48 hours) can reduce bacterial load and improve early pulmonary inflammatory response in a small model of acute pneumonia caused by Pseudomonas aeruginosa, without impairing the homeostasis of the small model 48 hours after infection.

 

Animal Model: Neutropenic murine thigh infection mode
Dosage: 400, 800 mg/kg
Administration: Subcutaneous injection (s.c.), every 6 h
Result: Reduced ESBL-producing Enterobacteriaceae. Showed that ceftolozane with tazobactam at a 2:1 ratio were significantly different than animals treated with ceftolozane alone.

    请依序添加每种溶剂: 10% DMSO  → 90% (20% SBE-β-CD in saline)

    以 1 mL 工作液为例,取 100 μL 25.0 mg/mL 的澄清 DMSO 储备液加到 900 μL 20% 的 SBE-β-CD 生理盐水水溶液中,混合均匀。
