Its metabolites participate in the synthesis of thymidine for incorporation into DNA or the synthesis of purines, as well as in the formation of methionine, which can be converted to the ubiquitous methyl donor, S-adenosylmethionine . THFA has also been used to study the activation of metabolite-binding riboswitches.

(6R, S) -5,6,7,8-Tetrahydrofolic Acid (hydrochloride) (0-200 μM; 3 days; Exposure to trihydrochloride in Adh5-/- DT40 cells exhibits cytotoxicity towards Adh5 and Fanconi anemia (FA) deficient cells, resulting in extensive DNA damage and chromosome accumulation interruption.
(6R, S) -5,6,7,8-Tetrahydrofolic Acid (hydrochloride) (0-100 μM; 16 hours; Treatment with trihydrochloride in Adh5-/- DT40 cells strongly promotes the dose-dependent formation of FANCD2 and ser139-H2AX focusing in Adh5-/- cells.
Due to uncontrolled activity, exposure to (6R, S) -5,6,7,8-Tetrahydrofolic Acid (hydrochloride) activates the action of DNA damage response (DDR) thymidylate synthase, leading to depletion of essential nucleotides and promoting repair through homologous recombination mechanisms.

(6R, S) -5,6,7,8-Tetrahydrofolic Acid (hydrochloride) (62.5 mg/kg; intraperitoneal injection; Every day; The treatment of Adh5-/- mice disrupts the hematopoietic function of hematopoietic cells, increases phosphorylation of ser139-H2AX, and reduces the survival rate of progenitor cells (HSPCs), indicating that excessive Tetrahydrofolic acid trihydrochloride may have mutagenic and genotoxic effects on bone marrow cells.
