ML-396 is a positive allosteric modulator of group III metabotropic glutamate receptors (mGluRs) with EC values of 108, 146, and 128 nM for mGluR4, mGluR7, and mGluR8, respectively, in calcium mobilization assays. ML-396 potentiates LSP4-2022-induced reductions in paired-pulse field excitatory post synaptic potential (fEPSP) slopes in mouse hippocampal slices. In vivo, ML-396 (30 mg/kg) restores hippocampal long-term potentiation, improves novel object recognition, contextual fear learning, and social memory, and decreases sleep apneas in the Mecp2 mouse model of Rett syndrome.

生物活性

ML-396 is a positive allosteric modulator of group III mGluRs. ML-396 inhibits mGluRs with EC50s of 125 nM, 146 nM, and 108 nM for mGluR4, mGluR7, and mGluR8, respectively in calcium mobilization assays. ML-396 reverses deficits in contextual fear memory, social recognition, and apneas in Rett syndrome (RTT) model mice.

体外研究

ML-396 (1 μM; 5 minutes) has an enhancing effect on the ramp reduction of field excitatory postsynaptic potentials (fEPSPs) induced by 30 μM LSP4-2022 on coronal sections containing hippocampus.

ML-396 (1 μM; 10 minutes) cannot change the slope of fEPSP alone, indicating that under stimulation conditions, mGlu7 does not have reinforcing activity in Mecp2^-/y sections.

体内研究

ML-396 (30 mg/kg; intraperitoneal injection; Once a day for a total of 17 days, in Mecp2^+/- mice, synaptic plasticity defects and learning and memory phenotypes were rescued, and the frequency of respiratory pauses was reduced. However, in Mecp2^+/+animals, this effect was not observed.

ML-396 (10 mg/kg; intraperitoneal injection; The plasma/brain partition coefficient of single dose in adult male Sprague Dawley rats is 1.67, and the predicted free brain concentration is approximately 40 nM.
